Product Specific Information

The BM8 monoclonal antibody reacts with mouse F4/80 antigen, an approximately 160 kDa surface receptor belonging to the EGF-TM7 family of proteins. It contains seven EGF-like domains, seven transmembrane sequences, and a cytoplasmic tail homologous to other TM7 family members.
F4/80 is expressed on most mature macrophages and serves as a key marker for this population. Other cell types, such as peritoneal eosinophils, Langerhans cells, and certain dendritic cell subsets, may also express F4/80. Expression begins early in myeloid development and can be upregulated by M-CSF stimulation in vitro. Some macrophage subsets, particularly in lymphoid tissues, may lack F4/80 expression.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes and bone marrow cells; ≤0.25 µg/test recommended. One test corresponds to staining in a 100 µL final volume (10⁵–10⁸ cells/test). Optimize antibody concentration for specific assays.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

F4/80 antigen is a 160 kDa cell surface glycoprotein, part of the EGF-TM7 family, sharing 68% amino acid identity with human EMR1 (EGF module-containing mucin-like hormone receptor 1). Expression varies with macrophage maturation and activation, and is found on Kupffer cells, Langerhans cells, microglia, peritoneal, pulmonary, thymic, and splenic red pulp macrophages. It may also be present on some dendritic cell subsets but is absent in macrophages of T cell zones.
Although ligands and biological roles are not fully defined, F4/80 is suggested to contribute to efferent CD8⁺ regulatory T cell generation.
