Target Information
TRIM5α is a 493 amino acid member of the tripartite motif (TRIM) protein family. TRIM proteins contain three zinc-binding domains (RING, B-box 2, and coiled-coil) and often form homomultimers to localize within different cell compartments. Some members, including TRIM5α, possess a C-terminal B30.2 (SPRY) domain and localize to the cytoplasm. TRIM5α mediates innate intracellular retroviral resistance, dependent on its C-terminal domain. The variable regions of the B30.2 domain form loops that may create a viral binding surface. TRIM5α trimerization is essential for its retroviral capsid affinity and antiviral activity. The linker region between the coiled-coil and B30.2 domains is required for trimerization. TRIM5α blocks infection after viral entry.
