E3 ligase 복합체의 기능적 특이성은 기질 인식 서브유닛에 따라 결정되며, CUL4B는 기질과 유비퀴틴 결합 효소의 위치를 조정하는 스캐폴드 단백질로 작용합니다.
방사선 유도 DNA 손상에 대한 히스톤 H2A, H3, H4 및 CDT1의 폴리유비퀴틴화를 매개하며, DDB2에 의해 UV 손상된 크로마틴으로 타깃팅됩니다.
Cyclin E의 유비퀴틴화를 조절하여 정상적인 G1 세포주기 진행에 필수적이며, mTOR 경로를 조절하여 세포 성장과 대사 조절에 관여합니다.
CUL4B의 mTORC1 매개 경로 조절은 26S proteasome 기능에 의존하며 MLST8과의 상호작용을 필요로 합니다.
