Target Information

Programmed death-ligand 2 (PD-L2), also known as B7-DC, is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for PD-1, an inhibitory immune checkpoint molecule that regulates T-cell activation and immune response. PD-L2 is primarily expressed in dendritic cells but can also be induced in other immune and non-immune cells depending on cytokine conditions such as IL-4, TNF-α, and IFN-γ. Compared to PD-L1, PD-L2 has a higher affinity for PD-1 (2–6 times greater).

PD-L2 not only inhibits T-cell activation but has also been shown to stimulate T-cell proliferation and cytokine production, even in PD-1-deficient cells, suggesting alternative receptor interactions. One such receptor is RGMb (repulsive guidance molecule b), which is involved in respiratory tolerance and BMP signaling. The human PD-L2 monomer consists of a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
