Product Specific Information
Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
GAD-65와 GAD-67은 group II decarboxylase family 단백질로, L-glutamic acid로부터 GABA(gamma-aminobutyric acid)를 생성하는 과정의 속도 제한 단계를 촉매합니다.
두 GAD 모두 뇌에 존재하지만, GAD-65는 신경 말단의 시냅스 소포막에, GAD-67은 세포 전체에 분포합니다.
GAD-67은 기본적인 GABA 합성을 담당하며, GABA 수요가 증가할 때 GAD-65가 일시적으로 활성화되어 합성을 돕습니다.
GAD-65의 결손은 GABA 신경전달에 장애를 초래할 수 있으며, GAD-67의 결손은 치명적입니다.
대체 스플라이싱으로 인해 GAD-67의 두 가지 isoform(GAD-67, GAD-25)이 존재합니다.
