Target Information
CIITA contains an acidic transcriptional activation domain, four leucine-rich repeats (LRRs), and a GTP-binding domain. It is located in the nucleus and acts as a positive regulator of class II major histocompatibility complex (MHC II) gene transcription, serving as the "master control factor" for these genes. CIITA binds GTP to facilitate its nuclear transport and functions as a coactivator through intrinsic acetyltransferase (AT) activity rather than direct DNA binding.
Mutations in CIITA are associated with:
- Bare lymphocyte syndrome type II (hereditary MHC class II deficiency)
- Increased susceptibility to rheumatoid arthritis
- Multiple sclerosis
- Possibly myocardial infarction
