Target Information
The ABL1 proto-oncogene encodes a cytoplasmic and nuclear protein tyrosine kinase involved in cell differentiation, division, adhesion, and stress response.
c-Abl activity is negatively regulated by its SH3 domain; deletion of this domain converts it into an oncogene.
Its DNA-binding activity is regulated by CDC2-mediated phosphorylation, indicating a role in the cell cycle.
In chronic myelogenous leukemia (CML) and some acute lymphoblastic leukemias, the c-Abl proto-oncogene undergoes a (9;22) chromosomal translocation, forming the Philadelphia chromosome through fusion with the Bcr gene.
The c-Abl oncogene was first identified as the transforming gene of Abelson murine leukemia virus (A-MuLV).
c-Abl also plays a role in DNA repair by activating proapoptotic pathways when DNA damage is irreparable.
