Product Specific Information
This F38-2E2 monoclonal antibody reacts with human CD366, also known as T cell immunoglobulin and mucin domain-containing protein 3 (TIM3) or HAVCR2.
CD366 is expressed on activated CD4+ T cell subsets (Th1, Th17, Treg), CD8+ T cells, monocytes, dendritic cells, and mast cells.
Galectin-9 is identified as the ligand for CD366. In humans, this receptor negatively regulates CD4+ T cells, influencing Th1- and Th17-related cytokine secretion.
CD366 is implicated in tolerance, autoimmune disease (e.g., multiple sclerosis), and HIV infection.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometric analysis of stimulated normal human peripheral blood cells.
Use at 5 µL (0.125 µg) per test (defined as staining a 100 µL cell sample).
Cell number range: 10⁵–10⁸ cells/test.
Excitation: 488–561 nm
Emission: 578 nm
Compatible Lasers: Blue, Green, Yellow-Green
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
TIM3 (HAVCR2) is a 281 amino acid, type I Th1-specific cell surface glycoprotein expressed on terminally differentiated CD4+ Th1 and CD8+ Tc1 cells.
TIM3 contains an IgV-like domain, a mucin-like domain, and a conserved tyrosine phosphorylation motif.
It is involved in macrophage activation, autoimmune disease induction, and immune tolerance regulation.
Pathological significance includes experimental autoimmune encephalomyelitis (EAE) and Th1-mediated immune response modulation.
