MAP4K4 (HGK)는 serine/threonine kinase로서 스트레스 활성화 MAPK 신호 전달 경로에 중요한 역할을 하며, TNF-alpha와 같은 사이토카인 및 환경적 스트레스에 대한 반응에 관여합니다. JUN N-terminal 경로의 상위 단계에서 작용하는 것으로 알려져 있으며, adapter 단백질 Nck의 SH3 도메인과 상호작용합니다. HGK는 CNH 조절 도메인을 통해 SPG3A의 N-말단 영역에 결합합니다. 모든 조직에서 발현되며, isoform 5는 뇌에서, isoform 4는 간, 골격근, 태반에서 더 풍부하게 발현됩니다.
