Target Information
Pyridoxal kinase (PDXK) converts vitamin B6 to pyridoxal-5-phosphate (PLP), an essential cofactor in amino acid and neurotransmitter metabolism. The PDXK gene encodes a 312-amino acid polypeptide exhibiting pyridoxal kinase activity. A major 1.5-kb transcript is expressed in all tested tissues and shows circadian oscillations. Expression in mouse liver and brain is regulated by PAR bZIP transcription factors (Dbp, Hlf, and Tef), which also oscillate. Mice lacking these factors show reduced brain PLP, serotonin, and dopamine levels, and are prone to spontaneous and audiogenic epilepsies.
