Target Information
FOXO1 (FKHR, Forkhead Box O1) is a transcription factor belonging to the forkhead homeotic gene family. It can act as a coactivator or corepressor of nuclear receptor activity through its LXXLL motif. FOXO1 association with PAX3 is implicated in alveolar rhabdomyosarcoma.
It plays a key role in autophagy, myogenic differentiation, and insulin signaling, regulating metabolic homeostasis under oxidative stress. FOXO1 binds to insulin response elements (IRE) and Daf-16 family binding elements (DBE), controlling redox balance, osteoblast numbers, and bone mass.
In hepatocytes, FOXO1 promotes gluconeogenesis by activating genes such as IGFBP1, G6PC, and PPCK1 together with PPARGC1A. It acts downstream of CDK1, PKB/AKT1, and MST1 to regulate cell death.
