The G-protein-coupled receptor (GPCR) superfamily comprises an estimated 600–1,000 members and represents the largest known class of molecular targets with proven therapeutic value.
The nociceptin receptor is a G-protein-coupled opioid receptor that functions as a receptor for the endogenous neuropeptide nociceptin. Ligand binding induces a conformational change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of downstream effectors.

Signaling through G proteins mediates inhibition of adenylate cyclase and calcium channel activities. Arrestins modulate G protein signaling and mediate activation of alternative pathways leading to MAP kinase activation. The receptor plays a role in modulating nociception, perception of pain, and regulation of locomotor activity by nociceptin.
