Target Information
Adrenomedullin (ADM) is a hypotensive peptide belonging to the peptide superfamily that includes calcitonin gene-related peptide (CGRP) and amylin. ADM was originally identified in the adrenal medulla and is highly expressed there. It is also produced by most contractile cells and is upregulated during sepsis and ischemia.
Three distinct receptors can bind ADM: ADM receptor (L1), RDC-1, and Calcitonin Receptor-Like Receptor (CRLR). CRLR associates with receptor activity-modifying proteins (RAMPs), which determine binding specificity.
- Co-expression with RAMP1 → CRLR binds CGRP
- Association with RAMP2 or 3 → CRLR binds ADM
The ADM receptor is a seven-transmembrane G protein-coupled receptor (GPCR) that mediates ADM signaling via G proteins activating adenylate cyclase. It is highly expressed in the heart, brain, skeletal muscle, immune system, adrenal gland, and liver.
