Target Information
Vacuolar-type H⁺-ATPase (V-ATPase) is a multisubunit enzyme responsible for acidification of eukaryotic intracellular organelles. It pumps protons against an electrochemical gradient, while F-ATPases reverse the process to synthesize ATP. The enzyme consists of a peripheral V1 domain (ATP hydrolysis) and an integral V0 domain (proton translocation).
The V1 domain includes nine subunits (A–H), and the V0 domain includes five subunits (a, d, c, c′, and c″).
The V-ATPase V1 B subunit exists as two isoforms. In the inner ear, the B1 isoform is essential for proton secretion and maintaining endolymph pH and auditory function. The gene encoding the human V-ATPase B1 isoform maps to chromosome 2cen–q13. Mutations in this gene cause distal renal tubular acidosis with sensorineural deafness. The B2 isoform is expressed in the kidney and is the only B isoform expressed in osteoclasts.
