Interleukin-12 (IL-12), also known as IL-12 p70, is a heterodimer composed of two subunits, IL-12A (p35) and IL-12B (p40), with a molecular weight of 70–75 kD.

IL-12 p70 is a secreted protein mainly produced and secreted by macrophages, dendritic cells, monocytes, neutrophils, and other antigen-presenting cells in response to antigenic stimulation. It plays an important role in the maturation, activation, proliferation, and immune regulation of immune cells.

As a T-cell stimulator, IL-12 p70 can effectively promote T-cell proliferation and function and participate in the differentiation of naïve T cells into Th1 cells. IL-12 p70 is also a cytotoxic lymphocyte maturation factor. In the tumor microenvironment, IL-12 p70 stimulates activated T cells and natural killer cells to release cytotoxic enzymes or secrete effector cytokines such as IFN-γ. These effector molecules are critical for tumor clearance.

검출 원리

본 키트는 Sandwich ELISA를 사용하여 사람 혈청 및 혈장 내 IL-12 p70의 양을 측정합니다.

1. ELISA 마이크로플레이트는 고친화성 anti-human IL-12 p70 항체로 미리 코팅되어 있습니다.

2. 표준물질 또는 시험 시료를 마이크로플레이트 웰에 첨가합니다.

3. 배양 후 시료 내 IL-12 p70이 웰에 코팅된 항체에 특이적으로 결합합니다.

4. 세척 후 biotin-labeled anti-human IL-12 p70 검출 항체를 첨가합니다.

5. 재배양 후 검출 항체가 웰에 결합된 IL-12 p70에 특이적으로 결합합니다.

6. Horseradish peroxidase-labeled streptavidin (Streptavidin-HRP)을 첨가하고 배양합니다.

7. 검출 항체의 biotin과 streptavidin이 강한 비공유 결합을 형성하여 다음과 같은 면역 복합체를 구성합니다.

   Coated antibody–human IL-12 p70 protein–detection antibody–Streptavidin-HRP

8. 다시 세척한 후 발색 기질 TMB를 첨가합니다.

9. HRP가 TMB 기질을 촉매하여 청색을 생성하며, 색의 강도는 시료 내 IL-12 p70 농도에 비례합니다.

10. Stop solution을 첨가하여 반응을 중지합니다.

11. 450 nm에서 흡광도를 측정하며, 570–630 nm를 참고 파장으로 사용할 수 있습니다.

12. 표준곡선을 작성하여 흡광도 값으로부터 시료 내 IL-12 p70 농도를 계산합니다.
