The monoclonal antibody eBio12A5 recognizes FR4 (Folate receptor 4, also known as FR delta or FBP3).
FR4 is a heavily glycosylated, 35 kD receptor for folic acid and the physiologic circulating form of N5-methyltetrahydrofolate.

Natural Treg cells express high levels of FR4 and, together with CD25, can be used to distinguish:

• Natural Treg (FR4^high CD25^high; high Foxp3)
• Central memory T cells (FR4^high CD25^low; high IL-2 production upon stimulation)
• Effector memory T cells (FR4^low CD25^high; secrete IFNγ and IL-17)

Co-staining studies indicate that epitopes recognized by eBioTH6 and eBio12A5 differ, allowing functional studies using both antibodies.

Target Information

FR4 is a heavily glycosylated 35 kD receptor for folic acid and N5-methyltetrahydrofolate.
Natural Tregs express high FR4 levels and can be distinguished from effector and memory T cells using FR4 and CD25 markers.
In vivo studies with eBioTH6 show decreased CD4⁺CD25⁺ cells and enhanced tumor response, leading to elimination of advanced-stage tumors.
