Target Information
GRP78 is a 78 kDa glucose-regulated protein belonging to the heat shock protein family (HSP70). It resides in the endoplasmic reticulum (ER) and transiently associates with newly synthesized secretory and membrane proteins. GRP78 also interacts with mutant and misfolded proteins, marking them for degradation.
The conserved sequence Lys-Asp-Glu-Leu (KDEL) at the C-terminus ensures ER protein retention via the KDEL receptor. GRP78 plays a key role in protein folding, degradation of misfolded proteins, and maintenance of cellular homeostasis, helping prevent apoptosis. It is a reliable biomarker for hypoglycemia and provides neuroprotection against glutamate and oxidative stress. Reduced GRP78 levels are observed in Alzheimer’s Disease and in cells with PS1 gene mutations. Induction of GRP78 is also linked to antitumor drug resistance.
