p16INK4a is a tumor suppressor protein that specifically inhibits CDK4/CDK6. It plays a critical role in the pathogenesis of various malignancies. Mutations or deletions of the p16INK4a gene are found in melanomas, gliomas, esophageal, pancreatic, lung, and urinary bladder carcinomas, and certain leukemias.
Expression of p16INK4a correlates strongly with HPV infection in head and neck squamous cell carcinomas (HNSCC). p16 status serves as an important prognostic indicator, with the p16-positive/HPV16-negative subgroup representing a distinct clinical category.
Target Information
The CDKN2A gene produces multiple transcript variants differing in their first exons. Two variants encode CDK4 inhibitors, while another transcript encodes an alternate open reading frame (ARF) that stabilizes the tumor suppressor p53 by sequestering MDM2.
Despite structural and functional differences, both CDK inhibitor isoforms and the ARF product contribute to cell cycle G1 regulation.
CDKN2A is frequently mutated or deleted in many tumors and is recognized as a key tumor suppressor gene.
