This product is diluted and ready-to-use.
A recommended positive control tissue is Glioma, though other tissues may also be suitable.
The primary antibody is intended for use by laboratory professionals in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissue during manual qualitative immunohistochemistry (IHC) testing. It is used after tumor diagnosis by conventional histopathology.
About ATRX:
Transcriptional regulator ATRX (ATP-dependent helicase ATRX, X-linked helicase II, XNP) is encoded by the ATRX gene. It contains an ATPase/helicase domain and belongs to the SWI/SNF family of chromatin remodeling proteins. ATRX is required for deposition of histone variant H3.3 at telomeres and other genomic repeats, maintaining silencing at these sites. ATRX mutations are common in CNS gliomas.
Usage Notes:
- Use with formalin-fixed, paraffin-embedded sections.
- Pretreatment with heat-induced or enzymatic epitope retrieval is recommended.
- IHC staining involves sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate, followed by interpretation under a light microscope.
Controls:
- A positive tissue control must be included in every staining procedure.
- Positive control tissues should show weak positive staining for optimal quality control.
- Internal or external negative controls should demonstrate absence of specific staining and help identify non-specific background staining.
- If specific staining occurs in negative controls, results with patient specimens are invalid.
Target Information
ATRX encodes a protein with an ATPase/helicase domain, belonging to the SWI/SNF chromatin remodeling family. Mutations cause X-linked mental retardation (XLMR) with alpha-thalassemia (ATRX syndrome), affecting DNA methylation patterns and gene expression. ATRX undergoes cell cycle-dependent phosphorylation, regulating nuclear matrix and chromatin association, and plays roles in gene regulation and chromosomal segregation. Multiple alternatively spliced variants exist.
