Invitrogen™ Dextran, Alexa Fluor™ 488; 10,000 MW, Anionic, Fixable
Labeled dextrans are hydrophilic polysaccharides most commonly used in microscopy studies to monitor cell division, track the movement of live cells, and to report the hydrodynamic properties of the cytoplasmic matrix. The labeled dextran is commonly introduced into the cells via microinjection.

High Manufacturing Standards of Molecular Probes™ Dextrans
Molecular Probes™ offers over 50 fluorescent and biotinylated dextran conjugates across multiple molecular weight ranges. Dextrans are hydrophilic polysaccharides with moderate-to-high molecular weight, excellent water solubility, and low toxicity. They are biologically inert due to their poly-(α-D-1,6-glucose) linkages, which resist cleavage by endogenous glycosidases.
Molecular Probes™ fluorescent dextrans are typically brighter and more negatively charged than comparable products. Rigorous purification removes unconjugated dye, and thin-layer chromatography ensures absence of low molecular weight contaminants.
A Wide Selection of Substituents and Molecular Weights
Dextrans are conjugated to biotin or various fluorophores, including seven Alexa Fluor™ dyes, and available in nominal molecular weights of 3,000; 10,000; 40,000; 70,000; 500,000; and 2,000,000 daltons.
Dextran Net Charge and Fixability
The succinimidyl coupling method produces neutral or anionic dextrans. Alexa Fluor™ 488 dextrans are typically anionic. Proprietary methods can add more negative groups to create “polyanionic” dextrans.
For applications requiring fixation, “lysine-fixable” versions are available. These dextrans contain lysine residues allowing aldehyde-mediated fixation for immunohistochemical or ultrastructural analysis. All 10,000 MW Alexa Fluor™ dextran conjugates are compatible with aldehyde-based fixatives.
Key Applications Using Labeled Dextrans
- Neuronal tracing (anterograde and retrograde) in live cells
- Cell lineage tracing
- Neuroanatomical tracing
- Studying intercellular communication (e.g., gap junctions, wound healing, embryonic development)
- Investigating vascular permeability and blood–brain barrier integrity
- Tracking endocytosis
- Monitoring acidification (pH-sensitive conjugates)
- Studying hydrodynamic properties of the cytoplasmic matrix
Solubility Information
Dextran conjugates are soluble in aqueous buffers up to 10 mg/mL. Solubility decreases with molecular weight:
- 3,000 MW: ~100 mg/mL
- 10,000 MW: ~50 mg/mL
- 70,000 MW: ~25 mg/mL
- 500,000–2,000,000 MW: 5–10 mg/mL
