Product Specific Information

The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by a subset of thymocytes and most mature T cells, and is upregulated upon T-cell stimulation. CD27 binds to CD70, playing a key role in T cell–B cell interactions.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.8 µg) per test in a final volume of 100 µL. Cell number: 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample to antibody cocktails before combining with cells.
    • For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures engineered for optimal fluorescent signatures with reduced spillover and spread.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily, including CD40 and CD30. It is primarily expressed on lymphocytes and plays a role in regulating cell proliferation and apoptosis. CD27 is found as a disulfide-linked homodimer on mature thymocytes, peripheral blood T cells, and a subset of B cells. Upon T-cell activation, CD27 expression increases, and soluble CD27 (sCD27) can be detected in plasma, urine, or spinal fluid—serving as a prognostic marker in B cell malignancies. CD27-binding protein (SIVA) is involved in CD27-mediated apoptosis.
