The Ambion™ Single Cell-to-CT™ Kit provides a complete validated workflow for gene expression analysis of samples containing 1–10 cells. Each kit includes reagents for sample preparation, reverse transcription, pre-amplification, and qPCR—optimized together in a simple workflow completed in only 5 steps.

Pre-optimized Protocol Ensures Success and Saves Time

The workflow begins with a 7-minute sample preparation step, where cells are lysed reproducibly with minimal processing into a solution compatible with downstream RT-PCR, without purification. Subsequent steps include:

1. Reverse transcription using Superscript™ RT
2. cDNA amplification with TaqMan™ PreAmp Master Mix
3. qPCR with TaqMan™ Gene Expression Master Mix

All steps are optimized for maximum sensitivity while maintaining the entire sample in the same well to prevent loss or dilution.

Single-Cell Sensitivity

The kit addresses challenges in sample prep, reverse transcription, and amplification to achieve robust single-cell gene expression analysis. Detection of single-cell equivalents demonstrates linearity and sensitivity compared to 100-cell controls, with excellent reproducibility.
