Target Information

Beta-catenin, an adherens junction (AJ) protein, is a key component of cell–cell adhesion structures. AJs are essential for epithelial layer formation and maintenance by regulating cell growth and adhesion. Beta-catenin interacts with the cytoplasmic domain of E-cadherin, linking it to alpha-catenin and anchoring the complex to the actin cytoskeleton.

It also associates with cytoskeletal complexes containing adenomatous polyposis coli protein and microtubules, participating in signaling pathways involving tyrosine kinases, phosphatases, and Wnt/Wingless. These interactions regulate morphogenesis and cell proliferation.

Beta-catenin is expressed in hair follicle cells, basal and peripheral matrix cells, and inner/outer root sheath cells. Aberrant beta-catenin activity has been implicated in pilomatrixoma, medulloblastoma, colorectal, ovarian, and other cancers. In the nucleus, it coactivates Lef/Tcf transcription factors that drive expression of genes such as cyclin D and c-myc, promoting tumorigenesis.
