Target Information

CtIP is a 125 kD protein originally identified as interacting with the transcription repressor CtBP via the PLDLS motif (CtBP-Interacting Protein), suggesting a role in transcription regulation.
CtIP also interacts with BRCA1 through its C-terminal BRCT domains, indicating a potential tumor suppressor function.
This CtIP–BRCA1 interaction can be disrupted by DNA-damaging agents such as UV or gamma irradiation.
Li et al. (Nature 406, 210–215, 2000) demonstrated that ATM phosphorylates CtIP at serine residues 664 and 745; mutation of these sites prevents BRCA1 dissociation from CtIP, maintaining repression of BRCA1-dependent GADD45 induction upon ionizing radiation.
