Target Information

Modification of target proteins by ubiquitin participates in a wide array of biological functions. Proteins destined for degradation or processing via the 26S proteasome are coupled to multiple copies of ubiquitin. Attachment of ubiquitin or ubiquitin-related molecules may also result in changes in subcellular distribution or modification of protein activity.

Deubiquitination is catalyzed by proteases called deubiquitinating enzymes, which fall into four distinct families:

  1. Ubiquitin C-terminal hydrolases
  2. Ubiquitin-specific processing proteases (USPs)
  3. OTU-domain ubiquitin-aldehyde-binding proteins
  4. Jab1/Pad1/MPN-domain-containing metallo-enzymes

USPs represent the most widespread and conserved deubiquitinating enzymes across evolution. They release ubiquitin from conjugated proteins and contain conserved catalytic domains (Cys and His boxes) with variable N- or C-terminal extensions that contribute to substrate recognition, localization, and protein-protein interactions.
