Target Information
Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and is essential for brain development. Mutations, rearrangements, and amplifications in the ALK gene have been observed in various tumors, including anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer.
Chromosomal rearrangements often result in a fusion gene between ALK (chromosome 2) and nucleophosmin (NPM, chromosome 5), producing the oncogenic NPM-ALK fusion protein.
ALK encodes a receptor tyrosine kinase with transmembrane and extracellular domains, which are absent in the NPM-ALK fusion product. It shows the greatest sequence similarity to LTK and plays a crucial role in neuronal development.
