Target Information
PUMA는 Bax 및 Noxa와 함께 p53의 전사적 표적 중 하나인 Bcl-2 패밀리의 친사멸 단백질입니다. PUMA 유전자는 BH3 도메인을 포함한 PUMA-a와 PUMA-b 두 단백질을 암호화합니다. 이 단백질들은 Bcl-2에 결합하고 미토콘드리아에 위치하며, p53 반응 시 시토크롬 c 방출과 세포사멸을 유도합니다. PUMA는 p53 유도 세포사멸의 직접적 매개체로 작용할 수 있으며, PUMA 발현 억제 시 p53에 의한 세포사멸 반응이 감소합니다. 이는 PUMA가 cytochrome c/Apaf-1 의존 경로를 통해 p53 유도 세포사멸을 매개함을 시사합니다.
