Product Specific Information

The M17/4 monoclonal antibody reacts with mouse CD11a, the 180 kDa integrin alpha L, also known as lymphocyte function associated antigen-1 (LFA-1) alpha chain. LFA-1 is formed by non-covalent association of CD11a with CD18 (integrin beta 2) and acts as an adhesion molecule involved in lymphocyte and granulocyte function. CD11a is expressed by all leukocytes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.4 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining. Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD11a (LFA-1α, ITGAL) forms the αLβ2 integrin complex with CD18. It mediates leukocyte adhesion via ICAM-1, -2, and -3 and participates in lymphocyte costimulatory signaling. CD11a expression and activation are key in immune cell interactions, migration, and T-cell function.
Blocking CD11a is a therapeutic strategy for inflammatory diseases; Efalizumab (Raptiva) is an example of anti-CD11a antibody therapy.
Two transcript variants encode different isoforms.
Associated diseases include benign cephalic histiocytosis and leukocyte adhesion deficiency.
