Product Specific Information

Immunohistochemistry (PFA fixed):

  • Incubate antibody for 30 min at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

HLA-G belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin).
The heavy chain is anchored in the membrane and is approximately 45 kDa. The gene contains 8 exons:

  • Exon 1: leader peptide
  • Exons 2–3: alpha1 and alpha2 domains (peptide-binding)
  • Exon 4: alpha3 domain
  • Exon 5: transmembrane region
  • Exon 6: cytoplasmic tail

HLA-G is expressed on fetal-derived placental cells.
HLA and MHC antibodies play a significant role in immunopeptidomics, aiding in neoantigen identification and characterization via high-performance liquid chromatography coupled with tandem mass spectrometry.
