The Human Syntenin-1 Simplex ProcartaPlex Kit measures Syntenin-1 protein and is designed to be combinable with other Simplex kits, allowing you to create your own multiplex panel utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits (e.g., not using a pre-configured multiplex panel), only one buffer kit (Cat. No. EPX010-10420-901) is required for each assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on the principles of a sandwich ELISA, using two highly specific antibodies binding to different epitopes of one protein to quantitate multiple protein targets simultaneously using a Luminex instrument.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single 25–50 μL sample from plasma, serum, cell culture supernatants, and other biological fluids.

The Luminex MagPlex superparamagnetic microsphere beads used in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique signatures identifiable by Luminex xMAP detection systems (Luminex 200, FLEXMAP 3D, and MAGPIX). Similar to a sandwich ELISA, matched antibody pairs identify the target protein. Each bead is labeled with antibodies specific to one protein, and bound proteins are detected with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE).

Unlike ELISA, the capture antibody in ProcartaPlex assays is conjugated to a magnetic bead and not adsorbed to the microplate well, allowing reagents to remain free-floating. The Luminex instrument uses two lasers—one to identify bead type and another to quantify RPE fluorescence proportional to protein concentration. This enables profiling of multiple targets using less sample and equivalent time compared to a traditional ELISA.

ProcartaPlex multiplex panels are available in multiple formats across six species (human, mouse, rat, nonhuman primate, porcine, and canine).
