Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) ubiquitously expressed in all nucleated hematopoietic cells, comprising approximately 10% of all surface proteins in lymphocytes. CD45 is absent on non-hematopoietic cell lines and tissues.
This glycoprotein is crucial in lymphocyte development and antigen signaling, serving as a regulator of Src-family kinases. CD45 protein exists as multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains.
CD45RA is an isoform with restricted expression among lymphoid cell subtypes. Isoforms differ in their ability to translocate into glycosphingolipid-enriched membrane domains, depending on cell type and physiological state.
CD45 regulates T- and B-cell antigen receptor signaling, suppresses JAK kinases to modulate cytokine receptor signaling, and promotes cell survival by influencing integrin-mediated signal transduction, DNA fragmentation during apoptosis, and immunological function modulation.
