Target Information
Ubiquitin-mediated proteolysis requires the transfer of ubiquitin (Ub) to lysine groups on selected cellular proteins, leading to degradation by the 26S proteasome. Ub-fusions are processed by Ub-specific proteases (UBps) or via the Ub-fusion degradation (UFD) pathway. The UFD1 protein, first identified in yeast (S. cerevisiae), has a human homolog known as UFD1L. UFD1 modulates degradation of Ub-fusions with amino acid substitutions at Gly76 through selective multiubiquitination. Mutations in UFD1 are linked to CATCH22 syndrome, characterized by cardiac defects, cleft palate, and hypocalcemia, indicating its role in developmental processes.
