Target Information
Cytokine receptor signaling is negatively regulated by the SOCS (Suppressor of Cytokine Signaling) family, which includes eight members: SOCS-1, 2, 3, 4, 5, 6, 7, and CIS. SOCS proteins contain an N-terminal region of variable length, a central SH2 domain, and a C-terminal SOCS box. These proteins function as part of a classical negative feedback loop that modulates cytokine signal transduction through a STAT-induced transcriptional mechanism. Transcription of SOCS genes occurs rapidly in response to cytokines, and each member inhibits signaling in distinct ways.
