Target Information
BACE1 (beta-secretase)는 알츠하이머병(AD)의 아밀로이드 베타 펩타이드(Abeta) 생성에 관여하는 주요 효소입니다. 일부 조기 발병형 가족성 AD는 APP(amyloid precursor protein)의 ‘Swedish’ 돌연변이로 인해 BACE1에 의한 절단이 증가하여 발생합니다. 이러한 유전적 및 병리학적 근거를 바탕으로 BACE1 및 기타 APP 절단 효소(예: gamma-secretase)를 억제하는 치료 전략이 연구되고 있습니다. APP는 먼저 beta-secretase에 의해 절단되어 용해성 단백질 유도체와 막 결합형 C99 단편을 생성하며, C99는 gamma-secretase의 기질이 되어 4 kDa의 amyloid-beta 펩타이드를 형성합니다. 이 펩타이드는 알츠하이머병 환자의 뇌에 침착됩니다. BACE1은 베타 부위 APP 절단 효소(beta-site APP cleaving enzyme)로서, Asp2로도 알려진 새로운 막관통형 아스파르트산 프로테아제입니다.
