Target Information
Metadherin (Metastasis adhesion protein), also known as MTDH or LYRIC, is a novel protein that localizes with tight junction proteins ZO-1 and occludin in polarized epithelial cells. It is recruited during tight junction maturation rather than acting as a structural component. Metadherin is overexpressed in breast cancer tissue and xenografts, while expression is low in normal breast tissue. It binds to lung vasculature, a common site of breast cancer metastasis, via a C-terminal extracellular segment. Blocking this domain or inhibiting metadherin with siRNA has been reported to reduce metastasis.
