Target Information

SUPT6H (Suppressor of Ty 6 Homolog) is a transcription elongation factor that binds histone H3 and plays a key role in regulating transcription elongation and mRNA processing. It enhances RNA polymerase II (RNAPII) elongation and is essential for activation by the HIV-1 Tat protein.
SUPT6H forms a complex with IWS1 and the CTD of RNAPII subunit RPB1 (POLR2A), recruiting mRNA export factors (ALYREF/THOC4, EXOSC10) and histone-modifying enzymes (e.g., SETD2) to ensure proper mRNA splicing, export, and elongation-coupled H3K36 methylation.
Through association with SETD1A, it regulates class-switch recombination and somatic hypermutation by forming H3K4me3 marks on AICDA loci, and promotes myogenic gene activation by facilitating H3K27me3 demethylation via KDM6A stabilization.
