Intact IgG appears on a non-reducing gel as a ~150 kDa band and upon reduction generates a ~25 kDa light chain band and a ~50 kDa heavy chain.

Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning the specific antibody DNA sequences from immunoreactive rabbits. Individual clones are screened to select the best candidates for production.

Target Information

IFN gamma (Interferon gamma, Type II interferon) is a macrophage activation factor and immune interferon produced primarily by T-lymphocytes and natural killer cells in response to antigens and cytokines. It is a dimeric glycoprotein consisting of two 146 amino acid subunits (~45 kDa homodimer).

On SDS-PAGE, IFN gamma appears as a combination of 25, 20, and minor 15.5 kDa bands due to differential glycosylation. Its biological activity is species-specific; human IFN gamma does not cross-react with mouse IFN gamma.

Functions include:

  • Antiviral and tumor antiproliferative activity
  • Induction of class I and II MHC
  • Macrophage activation
  • Enhanced immunoglobulin secretion by B lymphocytes

Activation occurs through IFN gamma receptor I and II, triggering the JAK-STAT pathway. IFN gamma shows ~40% sequence homology between human and mouse forms. It is upregulated by IL2, FGF basic, and EGF, and downregulated by vitamin D3 or DMN. Gene mutations are associated with aplastic anemia.
