Target Information
Human Rad18 (hRad18) is a homologue of S. pombe Rad18. In yeast, Rad6 and Rad18 play crucial roles in post-replication repair, filling gaps in daughter strands during replication of damaged DNA. The Rad6–Rad18 interaction is critical in the early steps of lesion bypass. In vivo, Rad18 forms a tight complex with Rad6 via a conserved ring-finger motif, binding to damaged sites through single-stranded DNA binding. This leads to degradation of stalled replication components via the ubiquitin-conjugating enzyme activity of Rad6.
Human RAD18 maps to chromosome 3p24–25, a region often deleted in lung, breast, ovary, and testis cancers. The gene encodes a 495 amino acid protein (~56 kDa) and is ubiquitously expressed across human tissues.
