Target Information
HtrA1 is a 51 kDa protein belonging to the highly conserved mammalian HtrA serine protease family. It is a human homolog of bacterial HtrA and includes a secretory signal sequence, IGFBP-binding domain, and a Kazal-type serine-protease inhibitor domain at the N-terminus, along with a trypsin-like serine/protease domain and a PDZ domain at the C-terminus.
HtrA1 regulates cell growth and inhibits signaling mediated by TGF-beta family members. It is upregulated in the placenta during pregnancy, playing roles in implantation and placental physiology. HtrA1 also regulates IGF availability by cleaving IGF-binding proteins, acts as a tumor suppressor, and is downregulated in ovarian cancers and melanomas. Its involvement in osteoarthritis suggests therapeutic potential. Northern blot analysis shows high expression in placenta.
